Indications
Indications of Orlistat are intended for the treatment of obesity and as a component of a weight management programme for overweight adults and children over 12 years old.
The clinical efficacy of Orlistat has not been established in the absence of confirmed efficacy and toxicity studies.
In patients with a body mass index (BMI) of 25 or more, Orlistat may be an alternative. In patients with a BMI of >30 kg/m2 or a history of gastrointestinal disorders, Orlistat may be a first-line therapy.
Dose
Orlistat is administered orally with a weight-bearing aid as a single oral dose. It is recommended to start at the lowest dose, with a target weight for weight loss, for which weight maintenance is advised by healthcare professionals. The initial recommended dose of 0.5 mg is then gradually increased to achieve a target weight of at least 15% of body weight. The recommended dose is usually increased to 0.5 mg or decreased to 0.5 mg. The total daily weight loss for the first 3 to 6 months is then gradually decreased until the target weight is achieved. If the target weight is less than 15% of body weight, the dose should be increased to 0.5 mg. If the target weight is greater than 15% of body weight and the dose is no longer 0.5 mg, the dose should be decreased to 0.5 mg. The maximum recommended dosing frequency is once per day.
The use of Orlistat has not been assessed in patients who are pregnant or breastfeeding.

Orlistat is a lipase inhibitor. It works by blocking the absorption of fat in the body.
Inhibitors of Hmg Fat
Inhibitors of Hmg Fat reduce the absorption of about half of the fat consumed in meals. When Orlistat is used as a first aid or weight-loss aid, it should be avoided.

Pregnancy and Breastfeeding
Pregnant or breastfeeding women should not use Orlistat. It is not known whether Orlistat passes into the breast milk in a dose-dependent manner. However, it is unlikely that Orlistat will pass into the breast milk in a dose-dependent manner in the future.

Driving and operating operate to an excessive degree. It is important to maintain good control of the ability to see a clearly defined line drawn across the counter.

Contraindications
Orlistat is contraindicated in patients with a history of gastrointestinal disorders, such as peptic ulcer disease, or when the drug is used concomitantly with a reduced-calorie diet.

Contingent management with OrlistatOrlistat works by preventing the absorption of about 50% of the fat in the diet, which is believed to be due to the increase in calorie intake caused by obesity. Without the absorption of the excess fat being absorbed, there is often no more fuel for the small intestine to use for energy production. Therefore, the abdominal fat decreases in value as the body adjusts to the changing calorie intake.
